A continuous cell viability assay (0–84 hours) was performed to evaluate mannose toxicity to various human and mouse cell lines including mutants of Kdn-associated metabolic enzymes. The mannose feeding concentration was set at 0–200 mM, and the same concentration of GlcNAc was used as a control for the hyperosmolarity effect. Results shown are for (A and B) HEK293A cells; (C and D) HUVECs; (E and F) <t>BJAB</t> K88 cells (GNE WT); (G and H) BJAB <t>K20</t> cells (GNE-KO); (I and J) PMI WT cells; and (K and L) PMI-KO cells. (L) PMI-KO cells were always maintained in mannose-supplemented (25 μM) media for their survival during GlcNAC feeding. Data are representative of 2 independent experiments.

The RNAhyb program predicts a binding site starting at 480 nucleotides after the stop codon. Upper: sequence of the target site at the 3'UTR of DEPTOR; Lower: sequence of miR‐155. The 5' and 3' ends are specified. (B) Confirmation of the binding site for miR‐155 with luciferase assay in <t>U2932</t> cells. Y‐axis: relative luciferase activity. X‐axis, from left to right: U2932 cells transfected with miR‐155 and DEPTOR, with mt‐miR‐155 and DEPTOR, with miR‐155 and mt DEPTOR, and with mt miR‐155 and mt DEPTOR (wt: wild type; mt mutant). (C) Induction of phosphorylated mTOR (p‐mTOR) by miR‐155 in BJAB cells. An EGFP+ vector is used to over‐express miR‐155. After transfection and sorting, >90% of the cells are EGFP+. Overexpression of miR‐155 reduced the level of DEPTOR with secondary increase in the level of p‐mTOR. (D) A model for induction of p‐mTOR by miR‐155 in gastric diffuse large B‐cell lymphoma resistant to H. pylori eradication therapy. In the mTOR complex, DEPTOR together with either RAPTOR in mTORC1 or RICTOR in mTORC2 inhibit the activity of mTOR. WNT7A inhibits cell proliferation in Jurkat cells . (A) Percentage of cell proliferation measured in Jurkat cells, Jurkat-pLVX (stable transduced with empty-vector), or Jurkat-pLVX-WNT7A cells after 48 h of culture. Percentages of cell proliferation were obtained by taking Optical density (OD) values of the Jurkat cells as 100% cell proliferation. (B) Leukemia-derived Jurkat cells and PBMC were exposed for 48 h to 3 μg/mL of recombinant human WNT7a (+rhWNT7a). Afterward, the percentage of cell proliferation was measured by employing WST-1 and setting the value of non-treated control cells (Jurkat and PBMC for their respective assays) at 440 nm as 100% of cell proliferation.

Restoration of WNT7A in K562-, BJAB-, and CEM-Tet-WNT7A cells inhibits their cell proliferation . Graphs of the left column depict the percentage of cell proliferation measured in K562-, BJAB-, and CEM-Tet-WNT7A cells after 24-h and 48-h incubation with Doxycycline (Doxy). The empty-vector cell lines (K562-, BJAB-, and CEM-Tet) were also included as controls. The percentage of cell proliferation was measured by adding WST-1 to the cell cultures for 2 h and by reading the absorbance of treated and untreated cells at 440 nm. The value in each cell line of non-treated cells after 24 or 48 h was setting as 100% of cell proliferation. Right column graphs illustrate the histograms of the flow-cytometric analysis done for apoptosis detection utilizing Annexin-V-FLUOS as markers in non-treated (filled curve) and 48-h Doxy-treated cells (open curve). Ordinate: number of cells; Abscissa: fluorescence intensities.
